The clinical manifestations include sudden, circular patchy hair loss on the scalp and other areas such as eyebrows, eyelashes, beard, and body hair, along with dotted depression of finger/toe nails ( 6). However, patients with moderate to severe alopecia areata (SALT score≥50%), especially those with alopecia totalis or universalis, require more effective, better tolerated, and safer alternative drugs ( 3– 5).ĪA is a degenerative disease that affects hair follicles and is characterized by inflammatory cell infiltration around lesion follicles. Clinical drug regimens mainly rely on intra-lesion or systemic corticosteroids, minoxidil, and methotrexate. Currently, there are no available drugs for permanent AA treatment. Persistent AA and its variants can lead to significant scalp hair loss, adversely impacting the patient’s quality of life and psychological well-being ( 2). Limitations: Few randomized controlled trials met the inclusion criteria and insufficiency of eligible data.Ĭonclusion: JAK inhibitors are effective in alopecia areata, although associated with an increased risk.Īlopecia areata (AA) is a commonly occurring autoimmune disorder, with a prevalence rate of 2% in the United States ( 1). Any adverse effects occurred in 921 of 1508 patients a total of 30 patients discontinued the trial owing to adverse reactions. Results: 6 randomized controlled trials with 1455 patients exhibited SALT 50 (odd ratio, 5.08 95% confidence interval, 3.49-7.38), SALT 90 (OR, 7.40 95% CI, 4.34-12.67) and change in SALT score (weighted mean difference, 5.55 95% CI, 2.60-8.50) compared to the placebo. We enrolled in randomized controlled trials and observational studies of applying JAK inhibitors in alopecia areata. Methods: Eligible studies in PubMed, Embase, Web of Science, and Clinical Trials up to May 30, 2022, were searched. Objective: Evaluate the specific efficacy and safety of JAK inhibitors in alopecia areata by systematic review and meta-analysis. Department of Dermatology, Second Affiliated Hospital, Zhejiang University School of Medicine, Hangzhou, Chinaīackground: JAK inhibitors treat various autoimmune diseases, but an updated systematic review in treating alopecia areata is currently lacking.
